I'd say the Golden Rule is to make sure that both town and the Mafia have roughly equal chances of winning. That means that as much as possible, you should reveal information only when needed, and avoid trying to influence the players' decisions.
There are lots of established power roles, like cops, doctors, and blockers. But sometimes you can get creative and make your own roles.
You can decide if the roles will be announced before the game starts or not. The people here on the Sims forum always like to announce the included roles before the game starts.
There's also a wiki for Mafia games. You can look for lots of game templates there. Maybe add a theme if you like. A lot of the setups there have been tested for balance, so you can at least be sure that it's fair to both the Mafia and town.
Normally, around 1/4 of the players are Mafia. Sometimes this isn't the case, although some other factor usually compensates for it. And there are games with multiple Mafia factions; it's even possible for them to kill each other.
There are lots of possibilities. And trial-and-error. But as long as you do your job as a host properly (and the players actually participate), people are sure to have fun.

I might do a themed U-Pick where you pick a favorite thing of yours, but I won't say what role is assigned to which pick. You'll just know that certain roles exist so that the Mafia can have a chance.

Fresh! You forgot that if not all roles in the setup are unique, or if there are more roles than players, or if the setup has no roles revealed at the start, then the mense can claim. Because then the Mafia can make claims of town roles, too.
